Cells are the basic units of our bodies. We can see parts of their internal structures with the microscope. However, more structures and components on the micron and even smaller scale and the actual functions in the cell taking place at the molecular level have been beyond direct observation.

In recent years, fluorescence protein biosensor technology has developed rapidly and opened possibilities to monitor the cell dynamics even on molecular level. However, the analysis methods have not kept up with the pace of the technology. The available methods are quite rigid and unsystematic and cannot make use of all the available information from the new sensors.

This work focuses on studying cell polarity, which is how the cell reacts internally to external mechanical loading, systematically using the potential of dynamic fluorescence imaging. It introduces clustering methods into fluorescence images processing, establishes an integrated pipeline to obtain polarity information at both cellular and subcellular scales with higher accuracy and reliability, and further provides an efficient tool to analyze different forms of polarity.

To sum up, with the developed tools, we can identify and study the intracellular pathways for force transmission as well as the resulting chemical changes in the proteins and hence understand better the cell polarity and its role in cell migration. The developed analysis tools can be easily extended to other polarity issues, promoting the application of fluorescence proteins related techniques in exploring cellular processes.
